⁹What are the Main Parts of a Light Microscope? | -Eyepiece Lens: Look through to see said Image Magnified
-Objective Lens: Magnifies the Image as Well - has different Zoom
-Stage: Helps the Slide
-Clip: Holds the Slide
-Handle: To Carry the Microscope with
-Lamp: To Shine Light - makes it easier to see the Image
-Focusing Knobs: Moves the Stage up and Down to make the Image Better [Focused] |
How can you Prepare a Specimen before you put it under a Microscope? | -Light has to let Through the Specimen. If its Thick then just get a Thin Slice via Cutting it
-Get a Clean Slide [Glass or Plastic] and use a Pipette to drop 1 Drop of Water or Mountant, which will Secure the Specimen in Place
-Using Tweezers, place your Specimen on the Slide
-Add a Drop of Stain, if Needed [Colourless or Transparent] Stains can Highlight Key Structures eg Eosin --> Cytoplasm
-Put a Cover Slip at 1 End of the Specimen, at an Angle with a Needle
-Lower the Slip and Press it down with said Needle so not Air Bubbles are Trapped |
How should you View a Specimen? | -Clip the Slide with your Specimen onto the Stage
-Use the Lowest Powered Magnified Objective Lens first
-Then, using your Focusing Knob, fiddle with it until you get an Image that is Focused
-If needed, then switch to a Higher Powered Magnified Objective Lens.
-When Done, make a Drawing of your Specimen |
How can you find out the Magnification of an Image -
1. If you know the Lens Used
2. If you DON'T know the Lens Used | 1. Total Magnification = Eyepiece Lens Magnification x Objective Lens Magnification
2. Magnification = Image Size / Real Size |
