T. brucei concentrates pentamidine by an energydependent, high-affi nity uptake system. [Note: Resistance is associated with inability to concentrate the drug.] Although its mechanism of action has not been defined, evidence exists that the drug binds to the parasite’s DNA and interferes with its synthesis of RNA, DNA, phospholipid, and protein.
